High Throughput Sequencing and Data Analysis
High throughput sequencing (e.g., 454 pyrosequencing and Solexa) is becoming a standard tool in many applications where large amount of sequences must be obtained at relatively low cost. Access and availability to the sequencer and expertise in data analysis are two key issues often limit researchers in accomplishing their goals related to sequencing. We provide services related to both 454 and Solexa. Using the 454 GSFLX, more than 100 million bases can be obtained per run (1 million bases per sample) at 200-400 basepairs in length. Applications for the 454 include an analyzed mutation in the context of the neighbouring genetic sequence – “Built-in Quality Control”; Pyrosequencing assays are mutation-tolerant. Unlike hybridisation-based assays, Pyrosequencing analysis generates a correct sequence regardless of the appearance of a new, unexpected mutation; delivers the "gold standard" of genetic analysis: the sequence itself; with Pyrosequencing technology, there is great flexibility in primer placement. Therefore it is easy to design a Pyrosequencing assay to analyze virtually any genetic marker; genome-wide miRNA analysis, and metagenomic analysis of complex biological samples. Metagenomic studies are performed with a well-designed set of universal primers covering more than 90% of all sequenced 16S rRNA genes.
